Abstract

In the digestive tract ischemia/reperfusion causes morphological changes in enteric neurons (LINDESTRÖM and EKBLAD, 2004; RIVERA et al., 2009; PAULINO et al, 2011), and in diseases such as acute mesenteric ischemia and Crohn's disease presenting as pathophysiological necrosis of the intestine. This project aims to study in ileum enteric nervous system of rats subjected to ischemia/reperfusion of ileal artery in periods of 4 hours, 24 hours and 1 week: a) distribution of P2X2 and P2X7 receptors in neuron and enteric glial cell, b) the proportion of neurons immunoreactive to anti-HUC/D (neuronal marker) showing coexpression with P2X2 and P2X7 receptors, c) the proportion of enteric glial cells immunorreactive to S-100/GFAP and coexpression with P2X2 and P2X7 receptors, d) the profiles and cell density of neurons and enteric glial cells immunoreactive for P2X2, P2X7, anti-HUC/D (neuron) and S-100/GFAP (glial cell) and f) analysis of effect of the antagonists pyridoxal-phosphate-6-azophenyl-2,4-disulfonic acid (PPADS/non-selective for the P2X) and Brilliant Blue G (BBG, a selective blocker for the P2X7 receptor). The analysis will be done in a fluorescence microscope and scanning confocal laser. (AU)
