Role of the brilliant blue green (BBG) antagonist on the P2X7 receptor and enteric nervous system of the ileum rats submitted to ischemia and reperfusion: morphofunctional study

Abstract

Abdominal pain can be consequent to several causes, among the various possibilities can be intestinal ischemia. In the digestive tract ischemia / reperfusion (I / Ri) causes morphological changes in enteric neurons. This work aims to analyze the effect of the antagonist Brilliant Blue Green (BBG) and the P2X7 receptor on the enteric nervous system of the ileum of rats subjected to I / Ri (ischemia and reperfusion). The intestinal ischemia will be obtained by the obstruction of blood flow ileal arteries by the time of 45 minutes followed by reperfusion for 4, 24 hours and 14 days. There is a group without reperfusion. The sham group means no occlusion of the ileal arteries. The P2X7 receptor antagonist, BBG is injected 1 hour after ischemia in the group of 24 h, and for 5 consecutive days in the group of 14 days. The tissue will be prepared by methods of double and triple immunohistochemical markers P2X7 receptor with Nitric Oxide Synthase (NOS), neurofilament (NF), acetylcholine transferase (CHAT), calretinin (CALR), Neuropeptide Y (NPY) and Vasoactive Intestinal Peptide (VIP) and the pan-neuronal anti-HuC / D. For qualitative and quantitative analysis will be used fluorescence microscopy and Confocal Laser Scanning. Other techniques will be used for analysis: the P2X7 receptor protein expression by Western blotting, the presence of neutrophils by histochemical technique of myeloperoxidase (MPO), apoptosis by TUNNEL Technical and functional activity of intestinal motility, the technique will be by organ bath. After, there will be statistical analysis of group data. (AU)
