Abstract

The present study aims to investigate and compare the immunohistochemical expression of beta-2-adrenergic receptor by epithelial cells of the oral mucosa and by malignant cells of the invasive front of the oral squamous cell carcinoma. Seventy five oral squamous cell carcinomas and their surgical margins obtained from patients diagnosed and treated at the Departamento de Cirurgia de Cabeça e Pescoço e Otorrinolaringologia of the A.C. Camargo Cancer Hospital, São Paulo, Brasil, from 1970 to 2000, will be analyzed for beta-2 adrenergic receptor expression by epithelial cells of the oral mucosa and by neoplastic cells, the clinical history, the location and extension of the tumor, the classification by the TNM-UICC system, the microscopic characteristics, the treatment and clinical evolution of the tumor. Ten microscopic fields of each surgical margin or tumor will be captured from a high resolution digital camera attached to a binocular optical microscope containing an 40X objective. The immunohistochemical expression of the beta-2 adrenergic receptor by epithelial cells from the surgical margins and by neoplastic cells from the invasive front tumor will be automatically analyzed based on the intensity of brown staining of the cells in each microscopic field. A program developed in MATLAB language that determines numerical values based on the number of pixels from the segmented areas (positively immunostained by the beta-2 adrenergic receptor) will be used. The association of the beta-2 adrenergic receptor by epithelial cells of the oral mucosa with the clinical, microscopic, treatment and clinical evolution of the tumor will be calculated by chi-square test or Fisher's exact test with significance level of 5%.
