Abstract Clavulanic acid is a potent beta-lactamase inhibitor obtained from Streptomyces sp. and used in association with beta-lactamic antibiotics to combat resistance. Currently, the industrial purification of Clavulanic acid is based on liquid-liquid extraction with organic solvents and chromatographic steps. Therefore, new purification alternatives offering economical, ease and ecological advantages are of great interest. Within this context, Aqueous Two Phase Micellar Systems (ATPMS) can be considered an interesting alternative to the conventional purification techniques. These systems are formed primarily by micellar solutions of surfactants, offering hydrophobic and hydrophilic environments and thus allowing selectivity on the partitioning of biomolecules. The main goal of this work is to study Clavulanic acid partitioning in a non-ionic ATPMS composed by the surfactant Triton X-100 in the presence of salt ((NH4)2SO4) or polymer (dextran sulfate). A better comprehension of this biomolecule behavior in the system will guide future studies of Clavulanic acid extraction directly from the fermentation broth. Initially, we will map out the coexistence curve for Triton X-100 in the presence of the salt and of the polymer. Based on the curves obtained and stability results already available, we will investigate Clavulanic acid partitioning at different surfactant concentrations in the presence of (NH4)2SO4. Following, the partitioning will be investigated in the presence of dextran sulfate. | |
