EFFECTS OF CHRONIC MORPHINE ADMINISTRATION IN WOUND HEALING OF SLEEP DEPRIVED MICE

Abstract

The skin is the largest organ in the body and performs a host of primordial functions that keep the body alive. Damage to this system unleashes mechanisms that characterize the repair process called healing. Morphine is a drug with immunosuppressant properties whose chronic use may lead to increased infection and delayed wound healing. As the physiological integrity of the skin is essential for healthy living, sleep is a biological phenomenon crucial to the quality of life. Thus, reduction of sleep time in many aspects appears to affect the composition and integrity of several systems, especially the immune system. In this sense, this project will evaluate the effects of chronic administration of morphine and lack of sleep in the healing of skin. Will be used adult hairless mice, males and females, divided into the following groups: control+saline (Control), morphine (M), paradoxical sleep deprivation+saline (SD), paradoxical sleep deprivation+morphine (SD+M), chronic sleep restriction+saline (SR) and chronic sleep restriction + morphine (SR+M). Morphine (10 mg/kg, subcutaneous) will be injected two times a day every 12 hours throughout the experimental protocol. The animals will be submitted to head injury with the aid of a punch biopsy of skin and assessment of healing will occur for 21 days. At the end of the proposed protocols, animals will be euthanized using the method of decapitation for blood collection and posterior dosages of corticosterone, cytokines and vascular endothelial growth factor. Still, will be collected heart, liver, kidney, prefrontal cortex and skin test the performance of single cell gel (comet test) to examine possible DNA damage.
