Abstract

The rational developement of metal complexes in pharmacology and medicine involves different areas of knowledge, especially on inorganic chemistry, biochemistry and pharmacology. Numerous vital processes are strictly dependent on the presence of metal ions. Also, the fact is that many diseases are diagnosed or treated with the aid of metal complexes. These are highlights of gadolinium and technetium complexes as contrast agents, as well as platinum complexes as antitumor agents. Thus, the investigation of new metal complexes becomes important and extremely relevant in the search for new drugs effective in treating various diseases and withal safe to the human body. This project deals with the synthesis of new metal complexes of platinum(II), paladium(II), silver(I) and gold(I) with rimantadine, a bioactive organic ligant which has antiviral activity. The 1-(1-adamantyl) ethylamine, known as rimantadine, is a derivative of adamantane and similar to amantadine, a commertially available pharmaceutical, being used in prophilaxis nd treatment of influenza A. The mecanism of antiviral activity of riamantadine is basead on blocking viral replication of two phases: the inhibition of uncoating of the virus and then maturation. It is intended to characterize both the ligand and the complexes synthesized with aid of the techniques: elemental analyssis, vibrational spectroscopy in the infrared, nuclear magnetic resonance 1H, 13C and 15N, mass spectrometry and thermal analysis. Studies of in vitro biological activity of the ligand and complexes will be made, especialy antibacterial and antiviral assays, with the purpose of checking their pharmacological potential.
