Abstract

This Project is aimed at developing biosensors to detect micro-organisms such as Escherichia coli, using the concept of an extended electronic tongue (e-tongue), in which the sensing units of the array comprise enzymes and polyelectrolytes in layer-by-layer (LbL) films. Novel film architectures will be developed with distinct polyelectrolytes and enzymes such as lysozyme, pepsin and tripsin, in order to seek synergy in the interactions among the components and the analytes. Film fabrication will be monitored with various experimental techniques, including UV-Vis absorption and fluorescence spectroscopies, FTIR spectroscopy and atomic force microscopy, which will also serve to characterize the film properties. The methods of detection will involve electrochemical techniques, e.g. voltammetry and amperometry, and electrical impedance spectroscopy, in measurements to be done in liquid samples in the static mode or in a flow injection analysis (FIA) system. Because we anticipate a large variability in the biological samples, the massive amounts of data generated in the electrochemical and electrical techniques will be analyzed with information visualization methods. This will allow a further step toward optimization of the sensitivity and selectivity of the biosensors. As for the scientific problem to be addressed, attempts will be made to correlate the film properties and data from the sensing experiments with the molecular-level interactions between film components and the analytes.
