Abstract

In Brazil, the ornamental species of flowers and plants market moves, annually, more than three billion dollars. The chrysanthemum (Dendranthema spp., Asteraceae) is considered one of the most valuable commercial ornamental, being produced in more than 100 municipalities in the State of São Paulo. Diseases induced by virus and viroids have been a serious problem for this crop in several countries, mainly by vegetative propagation, which facilitates the spread of theses pathogens. Viroids can cause serious damages to the culture, specially Chrysanthemum stunt viroid (CSVd, genus Pospiviroid, family Pospiviroidae), considered an EPPO A2 quarantine pest, can induce dwarf and retard flowering. In Brazil, there are few studies on these pathogens, with a single report, in 1990, of a possible occurrence of CSVd in chrysanthemum in the State of São Paulo. This project aims to: (i) identify and purify a Brazilian isolate of CSVd collected in Atibaia, São Paulo; (ii) clone a monomer of CSVd; and (iii) synthesize DNA and RNA probes (radioactive and non-radioactive) of negative polarity for CSVd detection by Northern- and dot-blot hybridization.
