Abstract

This project aims to evaluate the bone repair process at the bone / implant interface in rats with induced osteoporosis and treated with raloxifene or alendronate submitted to histometric, immunohistochemical, epifluorescence microscopy and biomechanical analysis (reverse torque). For this, 80 rats (Rattus norvegicus Albinus, Wistar), weighing 200 grams will be divided into three groups according to the analysis to be submitted, subdivided into four other subgroups according to -induced osteoporosis and drug treatment (SHAM BD = Rats that underwent sham surgery and were fed with a balanced diet; OVX WT = rats that underwent bilateral ovariectomy and fed with a low calcium diet (osteoporotic) without drug treatment; OVX RAL = rats submitted to bilateral ovariectomy and fed with a low calcium diet ( osteoporotic), treated with raloxifene; OVX ALE = rats submitted to bilateral ovariectomy and fed with a low calcium diet (osteoporotic), treated with alendronate sodium). Each animal will receive two implants, one in each tibial metaphysis. The implants that will be installed in the right tibia with will have a machined surface, and the left tibia will receive an implant with modified surface by double acid-etching. Euthanasia of the animals shall be carried out in the periods of 14, 42 and 60 days after implants installation, depending on the analysis to be made by means of an overdose of anesthesia. Euthanasia periods are determined in accordance with the analysis to be performed. For the histometric (stained with hematoxylin and eosin) and immunohistochemical analysis, 20 rats will be euthanized at 14 days and 20 rats at 42 days after implant placement. For epifluorescence microscopy analysis, 20 rats will be euthanized at 60 days postoperatively. And for the biomechanical analysis (reverse torque), 20 rats will be euthanized at 42 days postoperatively. All data will be submitted to a normality curve for determining the most appropriate test (parametric versus nonparametric). The significance level of 0.05 will be adopted for all tests.
