Separation and purification of dendrimeric prodrugs from 3-HYDROXYFLAVONE and intermediates by HPLC

Abstract

Infectious parasitoses considered neglected diseases represent a great health problem for many countries and areas. Drugs available in the therapeutics are, generally, toxics and do not have good efficacy. So, the discovery and design of new chemotherapeutic agents are extremely needed. In this context, researchers from LAPEN (Laboratory of Design and Synthesis of Chemotherapeutics Potentially Active in Neglected Diseases, in the Faculty of Pharmaceutical Sciences, University of Sao Paulo) designed the dendrimeric prodrug of 3-hydroxyflavone, potentially active in leishmaniasis and Chagas disease. However, additional effort is needed towards simplifying the synthesis strategy, as well as for the establishment of separation and purification methods for this derivative, with the goal of their effective application in dendrimers technology at LAPEN. This said, the objective of this project is to develop a methodology for separation and purification of 3-hydroxyflavone first generation dendrimer (G1) and also of its intermediaries, using High Pressure Liquid Chromatography, with UV and visible detection, HPLC-UV-vis, as analytical method. With this purpose, different phases and eluent composition will be studied, aiming at obtaining compound peaks completely separated in the chromatogram. Also, UV spectra of 3-hydroxyflavone dendrimer and of its intermediaries will be evaluated and, finally, the products will be purified by semi-preparative HPLC. (AU)
