Abstract

The microbial community of persistent endodontic infection and / or secondary is different from that found in teeth with pulp necrosis, predominantly Gram-positive. Such a community can have greater resistance to endodontic procedures. The objectives of this study are: a) identify bacteria isolated from the root canal of teeth with failed endodontic and periapical lesion via microbial culture method and genetic sequencing. b) quantify endotoxins and proinflammatory cytokines root canal before, after chemomechanical preparation (PQM) and after dressing. c) Correlate microorganisms detected and levels of endotoxin and cytokines with clinical and radiographic findings. Sampling will be conducted before and after the PQM after dressing of 20 root canals of teeth with endodontic failure, and asymptomatic periapical lesion. Samples will be diluted, plated and incubated. After the incubation the colonies are biochemically characterized by sequencing, and genetics. Endotoxin levels will be quantified in the root canal through the turbidimetric (LAL Pyrogent 5000). Levels of proinflammatory cytokines IL-1±, IL-1², TNF and PGE-2 will be quantified using enzyme linked immunosorbent assay (ELISA). The data will be tabulated on Excel and statistically analyzed by SPSS for Windows. (AU)
