Abstract

Diabetes mellitus (DM) is a metabolic disorder characterized by hyperglycemia resulting from defects in insulin secretion or action which causes disturbances in the metabolism of carbohydrates, lipids and proteins. The incidence of diabetes increases alarmingly and studies using medicinal plants that have anti diabetic effects have increased interest from researchers. Species of Myrcia genus are used in folk medicine for the treatment of diabetes. However, there are a few studies that postulate mechanisms of action. Data from our laboratory show that treatment with crude extract of M. bella caused a reduction in blood glucose, serum lipids, increased glycogen and protein expression of insulin signaling pathway proteins as IRS-1, PI3-K and AKT in liver. Thus, it is important to further investigation of the mechanisms that explain this hypoglycemic action. The animals will be divided into four groups: control saline (control animals treated with saline), control treated (control animals treated with extract of M. bella), Diabetic saline (animals induced to diabetes by administration of streptozotocin and treated with saline) and treated diabetic (animals induced to diabetes by administration of streptozotocin and treated with extract of M. bella). Gene and protein expressions of key enzymes involved in the processes of glycogenesis, glycogenolysis and gluconeogenesis in the liver, as glucokinase, glycogen synthase, glucose-6-phosphatase and phosphoenolpyruvate carboxykinase (PEPCK), besides expression of key proteins of the signaling cascade insulin in liver, adipose tissue and skeletal muscle, as IR, IRS-1, PI3-K, AKT and GLUT4 will be analyzed (AU)
