Abstract

At this point on discussion of the energy matrix of the country, cellulases are relevant for the hydrolysis of lignocellulosic materials aimed at the production of second generation ethanol from sugarcane bagasse. This production involves three steps: pretreatment of bagasse, enzymatic hydrolysis of cellulose and fermentation of pentoses, which require further research and scientific and technological development. The pretreatment processes are designed to destabilize the complex cellulose-hemicellulose-lignin removing hemicellulose and/or lignin and allowing access of cellulolytic enzymes to pulp for its saccharification. Ozonolysis is a chemical pretreatment of lignocellulosic biomass and its action is due to the exposure of the material to ozone, a powerful oxidizing agent. Due to the differences between electronic lignin and carbohydrates, the use of selective oxidation processes, such as those based on ozone, presents great possibilities of success. The depolymerization of carbohydrate to fermentable sugars can occur in two ways: chemical or enzymatic hydrolysis. Enzymatic hydrolysis has several advantages over the chemistry, among which we should highlight: low energy consumption and high substrate specificity by avoiding the production of undesirable compounds. The solid state fermentation is a process that provides high concentrations of enzymes that can be applied in hydrolysis step. However, this technique lacks scientific and technological knowledge which enables your application on a large scale for different combinations of microorganisms and substrates, with a focus on filamentous fungi as fermentative agents and agroindustrial solid wastes as substrates economical and environmentally friendly. In previous work of the research group, three fungi isolated were good producers of cellulases/hemicellulases, namely thermophiles Myceliophtora sp. and Thermoascus aurantiacus and mesophilic Trichoderma reesei. In this context, the aim of this project is to study the production of cellulolytic enzymes by the three fungi on substrates composed predominantly by sugar cane, and the application of these enzymes in the saccharification of sugar cane bagasse raw and pre-treated with ozone. The enzyme preparations are used for each fungus individually or as enzyme cocktails from the mixture of the three extracts. The project is part of the Thematic Programme FAPESP Bioenergy Research (BIOEN) and Paulista Center for Bioenergy Research and is being proposed by the research line Bioenergy, Treatment and Utilization of Wastes of the Post-Graduate Program on Chemistry from the Institute of Biosciences, Letters and Exact Sciences of São José do Rio Preto.
