Immunohistochemical analysis of femoral bone metabolism of rats submitted to alcohol consumption and/or estrogen deficiency

Abstract

The association of osteoporosis and consumption of alcohol has shown synergistic and deleterious effect on bone tissue. Osteoporosis directly related to estrogen deficiency causes increased osteoclast formation and decreased synthesis of osteoblasts, causing an imbalance in bone remodeling. Excessive consumption of alcohol is a major public health problem and also has direct relation with bone loss by decreasing bone turnover, decreased rates of resorption and also osteogenesis. Thus, the aim of this project will investigate possible alterations in bone metabolism in femurs of rats subjected to chronic alcoholism and estrogen deficiency induced by ovariectomy. Will be used 90 rats (Rattus norvegicus, Albinus variation, Wistar) with 3 months old. The animals will be divided into six different groups of equal number according to the type of diet and the presence or absence of hormones. Group 1: sham ovariectomized (sham), water and free diet, Group 2: Sham and daily doses of 20% of the calories in alcohol and group 3: Sham isocaloric and feeding groups associated with alcohol supplied by an aqueous solution of sucrose and Group 4: ovariectomy, water and solid diet free, group 5: ovariectomy and 20% calorie alcohol, Group 6: ovariectomy and feeding isocaloric. After 8 weeks of starting the diet, all animals are sacrificed by cardiac perfusion and femurs are removed. The femurs will be evaluated according to the reading of the slides stained with HE and also by immunohistochemical analysis, using markers RANKL, bone sialoprotein, osteocalcin and anti-PARP p 85. The values obtained are tested for variance ANOVA and Tukey's significance level of 5%. (AU)
