Abstract

The disease caused by Salmonella is one of the major health problem worldwide in terms of morbid and mortality. Among the Salmonella enterica serovars, the Enteritidis (S. Enteritidis) comprises strains that have specialized ability to colonize the reproductive tissue of poultry and contaminate internal components of eggs, which allowed S. Enteritidis to become the most common isolated serovar of Salmonella spp. causing human disease. Although the reasons that led to this emergence are uncertain, it is believed that an epidemical and more virulent clone has been inserted in the country through the exchange of chickens with other countries. The aim of this project is to comparatively analyze by phenotypical tests, transcriptome sequencing and genome sequencing Salmonella Enteritidis strains isolated in the pre and post-pandemic period in Brazil. For this, it will be studied 48 S. Enteritidis strains, being 24 strains isolated in the pre-pandemic period (1968-1992) and 24 strains isolated in the post-pandemic period (1993-2013) in several places of Brazil. The strains isolated in the pre and post-pandemic period will be compared regarding the invasion to human and chicken's epithelial cells, survival capability against phagocytosis by human and chicken macrophages, survival to the acid and oxidative stress and ability to grow in the egg albumen. The comparison of the expressed genes in each of the phenotypical tests described above will be performed for one strain of Salmonella Enteritidis isolated in the pre-pandemic period and one isolated in the post pandemic period that will present significative difference in the majority of the phenotypical tests, using transcriptome sequencing. Data of the differential expression of genes will be validated by RT-PCR. Moreover, the two selected strains will have their genome sequenced and the minimal lethal dose (DL50) will be determined. Together, the results should contribute for a better characterization of S. Enteritidis, as well as, it will provide information about the differences between strains isolated in the pre and post pandemic period in the country, which could collaborate to elucidate the reasons of the beginning of the pandemic in Brazil, as well as, may provide additional data to other studies about the emergence of the serovar Enteritidis as the most prevalent one in several countries worldwide.
