Abstract

In this project will be investigated antigen/antibody to PSA (Prostate Specific Antigen) with model membranes immobilized on nanostructured films. PSA is a glycoprotein composed of single chain synthesized by epithelial cells of the prostate to be degraded in the body assumes an inactive form and enters the bloodstream. Changes in the architecture of the prostate gland increases the concentration of PSA in the blood and its measurement, along with other tests, it is important for the early diagnosis of prostate cancer and thus increases the chances of curing the individual. Three topics will be addressed: i) immobilization of antigen in LbL films (Layer- by- Layer films), in order to study the specific antigen-antibody interaction by spectroscopic and electrochemical; ii) immobilization of the antibody in the same process described in previous item; iii) immobilization of antigens or antibodies in liposomes to check the increased sensitivity in response to anti - PSA or PSA. With respect to immobilization of antigens/antibodies will be produced LbL films containing such materials in the presence and absence of phospholipids, towards a better stability. The immobilization of the antigen/antibody in LbL films will be alternated with positively charged polymers, poly (allylamine hydrochloride) (PAH) or polyethyleneimine (PEI). The growth of the films will be monitored by UV-visible spectroscopy and fluorescence. The structural organization of antigens and antibodies will be analyzed by circular dichroism and their interaction will be studied by electrochemical measurements and Surface Plasmon Resonance (SPR).
