Osseointegration of titanium implants installed without primary stability in diabetic rats: influence of local release of aminoguanidine

Abstract

Allogenic and alloplastic bone substitutes have been used to improve the conditions of the surgical site and facilitate the process of osseointegration in cases of absence of primary stability. Among the alloplastic substitutes has highlighted the poligel polymer, such as chitosan, which stands out for its abundance, biocompatibility, biodegradability and low toxicity. Among the numerous biological properties described for chitosan, the antioxidant, antimicrobial, inhibiting the formation of biofilms, anti-inflammatory drug developer and healing are the most relevant to the dental field. The purpose of this study is the histologic evaluation of the bone healing in diabetic rat around implants associated with chitosan/aminoguanidine. Will be made 60 implants of titanium ASTM F67 (Grade 4), with dimensions of 2.1 x 2.8mm, CNC machined around STAR ® model SR-20RII, with power of 2.2 kW. The implants will be divided (n=30) into Group A (health rat) and Group B (diabetic rat). These groups will be divided again into GI (machined), GII (chitosan), and GIII (chitosan/aminoguanidin). Following are implanted in femurs of 60 male rats (250 grams), which will be anesthetized (Xylazine / Ketamine) and shaved on its hind legs straight. After incision, divulsion, access to the bone bed and punching over milling (twist drill with 2.5 mm in diameter) under irrigation with saline, the surgical beds will be filled with the material to each group and the implants installed. Following is sutured (4.0 wire). The animals will be sacrificed at 60 days postoperatively. The pieces will be processed in MMA (Stevenel's blue / Alizarin red S).
