Abstract

Contagious Agalactia (CA) is a disease that affects small ruminants. This disease is classified by the World Organization for Animal Health (OIE) as notifiable disease to have significant economic impact on sheep and goats. It cause reduction or suppression of milk production, fever, septicemia, mastitis, abortion, and severe arthritis. CA is an multi - etiological syndrome, in which four microorganisms were identified as causing disease, including: Mycoplasma agalactiae (historically considered the primary); Mycoplasma mycoides subsp. mycoides Large Colony; Mycoplasma capricolum subsp . capricolum; and M. putrefaciens. Despite progress in studies on therapeutic methods and prevention strategies remain ineffective in controlling the CA. Thus, the generation of new alternative that provides better protection against M. agalactiae infections are needed. Therefore, this project aims the identification, purification and characterization of antigenic proteins of M. agalactiae. DNA coding sequences (CDS) of M. agalactiae proteins will be analyzed using bioinformatics techniques (SignalP version 4.1, TMHMM version 2.0, IEDB Analysis Resource). CDS will be selected from the surface proteins, secreted proteins or pathogenesis related proteins possessing antibody epitope prediction. The antigenic proteins of M. agalactiae will be cloned and expressed in Escherichia coli as a heterologous system. After cloning and purification to obtain the selected proteins, these will be evaluated for its antigenicity by using sera of goats naturally infected with M. agalactiae. (AU)
