Evaluation of bone healing in bone/implant interface in pinealectomized rats: histologicc, histometric, immunohistochemical, biomechanical and microCT analysis

Abstract

Currently, dentistry researches in rehabilitation with dental implants have been focusing in experimental studies and clinical trials, due to the accessibility and popularity of implant treatment. Melatonin is a hormone produced by the pineal gland, which can interfere with bone healing in several ways, by modulating inflammatory processes, the formation of collagen fibrils, time of osteoblast differentiation and also oxidative stress. Objective: This project intends to investigate the cellular changes and possible complications that may occur along the interface (bone / implant) in pinealectomized rats. The hypothesis of this study is that the peri-implant bone healing of pinealectomized rats is different from the control group. Hormone replacement melatonin could regulate this change. We believe the results of this study may emphasize the role of melatonin participating in the biological responses that manage bone metabolism, especially in night workers, who lose the ability of circadian release of melatonin by the pineal gland. Material and Methods: a total of 36 male adult rats (Rattus norvegicus Albinus, Wistar), will be divided into three groups (n = 12): control (CTRL), pinealectomized without melatonin (PNX), and pinealectomized with use of melatonin (PNXm). Animals will be subjected to the 1st surgical procedure (pinealectomy) except the CONTR group. 30 days after PNX , in the 2nd surgical procedure, all animals will receive an implant in each tibia. The animals of the PNX group will receive vehicle solution by gavage and rats of PNXm group will receive melatonin by gavage once a day until euthanasia at 42 days after implant placement. The three groups (n = 6 tibia for analysis) will be submitted to histological, histometric, imunohistochemic and biomechanical analysis. Quantitative values will be collected for each hystometric (ELCOI, Mr ELCOI, ELCOI re AON) and biomechanical (reverse torque) parameter, the data will be then tabulated to be subjected to statistical analysis.
