Abstract

The aim of this study will be to investigate the bone metabolism dynamics of bone / implant interface in SHR rats that will have the implant placement and analyze what the influences that losartan treatment may lead to peri-implant bone. 96 male rats will be used, 48 (SHR) and normotensive 48 (Wistar) will be performed surgery to install an implant in a tibia per animal. The analysis will be divided into groups: Group A, the tibia (n = 8) will be used for biomechanical analysis (reverse torque), and then after implant removed, the harvested tibia will be submitted to the Real time PCR analysis. Group B, the tibia (n = 8) will be used for MicroCT analysis and after performing the CT scan these tibia shall be submitted to the inclusion process in light-curing resin for making the slides the calcified cutting system EXAKT the slides will be used analysis by confocal laser microscopy, to analyse the bone dynamics at 14 and 42 days marked by the application of fluorochromes (calcein/alizarin red); for histometric analysis, the images taken by confocal microscope will be submitted to the precipitation area measurements of fluorochromes (Calcein and Alizarin) in the peri-implant region, and then the same will be subjected to staining Stevenel's blue and alizarin red, to measure linear extension contact bone/implant and newly formed bone area. Group C, the tibia (n = 8), shall be submitted to decalcification process for paraffin embedding, and the slides with semi-serial sections will be mounted where the odd will be used for immunohistochemical analysis with antibodies VEGF, OPG, RANKL and OC; tibia will be used for histological analysis, where the quality of the bone tissue maturation, inflammation and cell morphology will be assessed. All quantitative results will be statistically analyzed adopting a 5% significance level. (AU)
