Analysis of the biological responses involved in bone metabolism in spontaneously hipertensive rats non treated and treated with losartan: a immunohistochemistry and histomorphometric study

Abstract

The systemic arterial hipertension is one of the principal health problems of the population because of your high prevalence. Hipertensive individuals presents an anormal calcium metabolism, resulting in lower concentrations of this mineral, of D vitamin and magnesium and high concentrations of parathyreoid hormone, that may leads to a worse quality of bone tissue in an hypertensive patient, reducing the capacity of bone formation in situations as alveolar bone healing after the dental extraction. Losartan consists on an anti-hipertensive drug that has been used in the therapeutics of a great part of hypertensive patients.Therefore, the present project has the aim to evaluate the biological responses involved in the bone repair process after the dental extraction and after the realization of bone grafts in the model of spontaneously hypertensive rats that received or not the losartan treatment. The animals will be divided in 4 experimental groups: control, spontaneously hypertensive, control treated with losartan and spontaneously hypertensive treated with losartan. The animals will be submitted to the surgeries of upper right incisive extraction or to the installation of bone graft calvaria installed in the mandible region. The experimental periods will be of 7, 14 and 28 days after the surgeries.The evaluations will be performed through the histometric analysis of the slices included in paraffin and stained with hematoxiline and eosine. Another analysis will be through the immunohistochemistry reactions in order to label the bone matrix proteins involved with the responses of bone formation and resorption. This experiments will be also performed in the slices included in paraffin.The obtained results will be submitted to statistical analysis, stablishing comparisons between the different experimental periods as well as the different experimental groups.
