Abstract

The search for natural compounds in the presence of biological activities that assist the physiological functions in organisms have driven research by the pharmaceutical industry and especially food. In this sense, bioactive peptides have a high potential for use in the food industry, especially for the development of functional foods and beverages. Consequently, they have become the stimulus by the industry and academia to develop techniques and processes with different sources for the proteolytic release of these substances. Hydrolysis of casein by specific microbial proteases such as filamentous fungi results in the release of various peptides with different biological activities. Thus, screening and evaluation of these peptides for their application, may represent an important contribution to the functional food sector. Therefore, the aim of this study is to evaluate the antioxidant, antimicrobial and antihypertensive activity of peptides released by hydrolysis of caprine casein using proteases produced by filamentous fungus previously selected. For this purpose, will be initially evaluated the potential hydrolysis of five filamentous fungi producers of proteases selected from a database of 70 species, according to the evaluation of the biological activities of their hydrolysates. Subsequently, the fungus rated "GRAS" (Generally Regarded the safe) to present in their hydrolyzed the best results for the antihypertensive activity, will be selected for the optimization of protease production, purification and partial enzymatic hydrolysis of casein. The peptides released shall be purified, identified, evaluated for biological activities proposed and characterized molecularly. Thus, separation and purification techniques such as electrophoresis and high performance liquid chromatography using iodines arrangement coupled to a mass spectrometer detector of the type LC-MS/MS will be employed for both reports in the literature of these biomolecules, and with the aim of enhance the achievement of bioactive peptides.
