Effects of experimental periodontal disease induced by Aggregatibacter actinomycetemcomitans and Porphyromonas gingivalis in the fetal-placental unit and systemic inflammatory markers

Abstract

Epidemiological studies had shown that periodontitis were associated with low weight and premature birth. However, little is known about the mechanisms involved in this association. The complex microbiota associated with periodontitis includes Porphyromonas gingivalis and Aggregatibacter actinomycetemcomitans, which are considered among the main periodontopathogens. Although these organisms are able to induce distinct host responses, their potential in inducing systemic alterations has not been compared. In a pilot study, we observed that the oral infection with P. gingivalis in a mouse experimental model resulted in alveolar bone loss, disseminated infection, alteration in cytokines profile and reduction in fetal weight. However, due to the low number of studied animals, these conclusions were not definitive. This study aims to test the hypothesis that differences in the oral microbiota may have lead to differences in the potential to induce gestational alterations. The effect of experimental periodontal diseases induced by P. gingivalis strain HW24 D-1 and A. actinomycetemcomitans clone JP2 on the gestational pattern and inflammatory markers will be evaluated in C57BL6 female mice. The alveolar bone loss will be determined by microtomography (microCT). The amount of colonizing pathogens will be determined by qPCR in oral biofilm, gingival tissue, blood, serum, liver, linfonodes, gut, feces, fetus, placenta and amniotic liquid samples. The effect of the infection will be evaluated by determining the maternal weight gain, weight and number of fetus and fetal resorption. The level of the inflammatory markers IL1-², IL-2, IL-6, IL-5, IL-8, IL-10, IL-17, IL-18, TNF-±, GM-CSF, IFN-³, MIP-1, MCP-1, PGE-2 and RANKL will be determined by ELISA in serum, placenta and gingival tissue.
