Abstract

Osteosarcoma is the most common pediatric primary bone cancer. The treatment is based on surgical resection, followed by neoadjuvant and adjuvant multimodal therapy. However, around 80% of patients that present metastatic events at diagnosis have a poor overall survival rates. Lungs and other bones are the main affected organs and there still are no standard treatment for these cases. Therefore, a better understanding of metastatic process has become primordial, as well as the search for new therapeutic targets. The major pathway related with the invasion and migration by neoplastic cells involves the Rho family GTPases, whose principal effectors are the ROCK1 and ROCK2 kinases. These proteins are responsible for cell migration through cytoskeleton control and have been reported to be overexpressed in a wide range of neoplasms, including osteosarcoma. Moreover, high levels of ROCKs have been associated with poor prognosis. Recently, studies have pointed microRNAs deregulation in tumorigenesis process. The hypoexpression of some microRNAs has been related with ROCK kinases hyperexpression, therefore they may also be involved in the metastatic process. Thus, the aim of this project is analyze ROCKs kinases and microRNAs expressions in tumor samples and perform functional assays after induction or repression of associated microRNAs. (AU)
