Effect of protease inhibitors on collagen degradation potential from sound, sclerotic and caries-affected dentin

Abstract

The purpose of the study is to evaluate, through the tests of ultimate tensile strength and nanoindentation, the influence of proteases inhibitors on the collagen degradation from sound, sclerotic and caries-affected dentin. Thirty-nine human molars will be used, 13 for each dentin condition. Three slices will be obtained from each tooth. For the ultimate tensile strength, the teeth will be sectioned perpendicular to the long axis, worn in an hourglass shape and divided into 3 groups according to the protease inhibitor used: Group 1: artificial saliva (control group); Group 2: 2% chlorhexidine; Group 3: 0.5% EGCG (epigallocatechin gallate). After incubation, the samples will be subjected to enzymatic degradation (collagenase from Clostridium histolyticum), and then be submitted to the ultimate tensile strength test (MPa). The mechanical properties of dentin nanohardness and elastic modulus will be measured in digital nanoindenter under 1000¼N load. Data of ultimate tensile strength and mechanical properties of nanohardness and elastic modulus of collagen will be subjected to statistical analysis to verify the significance between the means (± = 0.05).
