Abstract

The antimicrobial peptides (AMPs) are defence molecules found in different organisms. Besides its antimicrobial activities, it has been shown that they possess potential antitumor activity. Our group described the ß-hairpin AMPs (protegrin, tachyplesin, polyphemusin and gomesin) cytotoxicity as well as some mechanisms triggered by them related to cell death of tumor cell lines such as K562, B16 and CHO (Chinese hamster ovary). We described that these peptides can cause direct membrane disruption in high concentrations while, in lower concentration, triggering different types of cell death after entry into tumor cells. Currently, we are extending the studies on ß-hairpin AMPs (protegrin, tachyplesin, polyphemusin and gomesin) and their anti-tumor activity concerning membrane interaction and organelle targeting by PHI (phototermal heterodyne image) microscopy and morphological, molecular and biochemical events of cell death. PHI microscopy is a technique that permits to follow a single molecule tracking such as AMPs. Thus, the use of PHI microscopy will allow us to monitor the entry of AMPs across cell membrane and follow these AMPs inside of cell determinate organelle targeting. Additionally, in the end of the project we also aim to analyze all data with computational mathematic multiparametric analysis, such as Bayesian nets, in order to evaluate all the pathways in an integrated way.
