Abstract

Ameloblastoma is an odontogenic tumour of slow growth, but locally infiltrative, preferentially affecting the posterior region of the mandible. Its malignant counterpart, the ameloblastic carcinoma, may arise de novo or develop from a previous ameloblastoma, being associated with a high rate of recurrence and metastases to lymph nodes and distant sites. Although rare, both lesions represent the most common benign and malignant odontogenic tumors diagnosed in most of the main Oral Pathology services. The molecular mechanisms involved in the pathogenesis of these neoplasms are poorly understood, and despite alterations in DNA repair system, in the regulation of apoptotic activity and cell proliferation favor the development of different human cancers, their importance in the development of ameloblastoma and ameloblastic carcinoma demands additional studies. Thus, the aim of this project is to analyze the expression of DNA repair proteins hMutS and hMutL, as well as the anti-apoptotic marker Bcl-2 and cell proliferation factor Ki-67 in ameloblastomas and ameloblastic carcinomas, determining the importance of these molecules for the development and clinical behavior of these tumours. For this, immunohistochemical reactions will be performed on 10 samples of normal tooth germs, 70 cases of ameloblastoma and 10 cases of ameloblastic carcinoma. The results will be quantified with the use of digital algorithms and correlated with clinicopathological parameters obtained from patients' medical records. By the end of this research it is expected to identify different protein expression patterns between the groups analyzed, leading to a better understanding of the pathogenesis of the lesions and assisting with the prognostic and diagnostic determination of these neoplasms.
