Abstract

Yeasts adapted to industrial conditions are considered appropriate platforms for the development of commercial strains applied to the production of chemical derivatives of renewable raw materials, in particular biofuels. However, understanding genetic mechanisms related to robustness or combine industrially relevant features for crossings require the collection and analysis of a large number of segregating. In Saccharomyces cerevisiae, obtaining haploid typically occurs by manual dissection of tetrads, an extremely laborious procedure depending on the number of haploid that is required. In this context, the implementation of techniques forhigh-throughput tetrad dissection has great impact on reducing the time necessary for the study or development of new strains. Therefore, it is intended to implement a method of analysis and dissecting tetrads based on flow cytometry and apply it to the study of genes relating to thermotolerance observed in industrial strains.
