Abstract

Evidence has shown that the same mutations often produce different phenotypes among genetically distinct individuals. These background effects are caused by modifier polymorphisms that segregate within species and alter the effects of mutations. In unpublished work, the Ehrenreich group at the University of Southern California has shown that genes involved in the regulation of histone acetylation are particularly likely to show background effects relative to other classes of genes. These genes play conserved roles in chromatin regulation and transcription from yeast to humans. Thus, understanding background effects involving these genes in yeast may shed light on how background effects occur in other species of clinical and industrial relevance. During my fellowship, I will go to the Ehrenreich lab to help characterize the genetic and molecular basis of background effects in histone acetylation. Specifically, I will use high-throughput CRISPR/Cas9-mediated genetic engineering and statistically powerful linkage mapping to dissect background effects in histone acetylation to causal genes and genetic variants. My work should produce new, fundamental insights into the mechanisms that cause background effects in eukaryotes.
