Evaluation of tissue recation caused by two different proportions of MTA flow after implantation in rat subcutaneous

Abstract

It was recently launched MTAFlow® an MTA-based material that was developed to easily acquire proper consistency for a variety of clinical applications. However, until now, there are no studies evaluating the properties of this material. Thus, this study aims to evaluate tissue response MTAFlow®, comparing it to Biodentine®, after implantation in the subcutaneous tissue of rats. For this, morphological and morphometrical analysis of the capsule adjacent to the implants with the materials will be made. Forty rats will be divided according to the material tested and experimental period. The materials will be mixed and inserted into polyethylene tubes. The MTA Flow® will be mixed in two different ratios following the manufacturer's instructions: 0.19 g of powder for 1 drop of gel and 0.06 g of powder for 1 drop of gel. The Biodentine® will be mixed according to the manufacturer's instructions. Then, the animals will be anesthetized and the tubes will be implanted in the subcutaneous tissue; empty polyethylene tubes will be used as control. After periods of 7, 15, 30 and 60 days, the animals will be euthanized and the tissues surrounding the polyethylene tubes will be removed and subjected to processing for paraffin embedding. Sections will be adhered to glass slides and stained with hematoxylin-eosin and Masson's trichrome. In the morphometrical analysis, the number of inflammatory cells and fibroblasts per mm2 will be quantified; the thickness of the capsule will be also measured. Other sections will be used for quantification of collagen in the capsule with the picrosirius technique associated with the analysis under the microscope with polarized light. All data will be subjected to ANOVA and Tukey test (p d 0.05). (AU)
