Abstract

This project has goals of applied order and of basic science. The first one is to establish an induction protocol for the final maturation and ovulation of lambari females (Astyanax altiparanae) using salmon GnRH analogue (sGnRHa). The second is to understand the hormonal mechanism in this species, which is considered a good model for breeding studies for other tropical species. Six equidistant levels of salmon GnRH analogue were tested, in the 0 to 160 ¼g kg-1 range and a positive control (6 mg EHC kg-1). For each treatment will be used five 10L experimental units, where four females are allocated (one for each collection time). Five collections will be performed at different times during the experiment: before (T0), three after the hormonal induction (T1), (T2) and (T3) and after spawning (T4), when the five remaining animals of each experimental unit will be collected. At each time, blood will be collected for analysis of the plasma hormone profile of estradiol, 17±-20²-dihydroxy-4-pregnen-3-one and prostaglandin F2±. In addition, gonads will be collected for the histological analysis of the presence of oocytes in the stage of germinal vesicle breakdown (GVBD) and post-ovulatory follicles after the time for ovulation and correlate with plasma hormone levels; and pituitary gland, for analysis of the gene expression of the beta subunits of lh. Fertility, fertilization and hatching rates and viability of the larvae, as well as gonads for the analysis of the percentage of post-ovulatory follicles and oocytes in GVBD will be collected of the five females that will remain until spawning.
