Abstract

The cells of apical papila (CPA) have direct influence in the root formation during dental development, acting as a source of dentin-synthesizing cells. If a pulpal pathological process is installed before the end of rhizogeneses, there is the possibility of pulpal necrosis and interruption of root formation. Although this cause/effect relationship is known, the molecular mechanisms involved in the process of rhizogeneses interruption and its relationship with CPA have not been clarified. Among the mediators potencially involved in this process, the endocannabinoids (eCBs) can be mentioned, which act in the modulation of several cellular activities. The aim of this study is to elucidate the role of eCBs in the molecular mechanisms involved with phenotypic and functional alterations of CPA in vitro. CPA cultures will be established and characterized from healthy immature third molars. Cells will be subjected to cellular viability assay through exposure to increasing concentrations of lipopolysaccharide (LPS). CPA will be challenged with LPS at proliferation (MP) and differentiation medium (MD) to detect the production of eCBs enzymatic system and expression of their receptors (CB1 and CB2). The influence of these receptors on the cellular modulation will be analyzed by means of pharmocological blockade. The role of CB1 and CB2 in cell differentiation and modulation of inflammatory bone resorption induced by LPS will be evaluated. Statistical analysis will be performed using one-way analysis of variance (ANOVA) followed by Tukey's post-test or Student's test. Significant values will be set at p<0,05. (AU)
