Abstract
The endodontic treatment of immature teeth is considered one of the greatest challenges of contemporary Endodontics. Despite the recent advances in this area of Dentistry, the molecular mechanisms involved with interruption of root formation resulting from dental caries or trauma are still unknown. Therefore, the aim of this study is to elucidate the role of inflammatory mediators in functional alterations of apical papilla cells (CPA). In vitro cultures of CPA will be established from third molars with incomplete root formation. The cells will be subjected to stimulum with increasing concentrations of lipopolysaccharide (LPS) for cellular viability assay. Then, CPA will be challenged with LPS at proliferation (MP) or differentiation medium (MD) for detection of prostaglandin E2 (PGE2), endocanabinoids or angiotensin II (Ang II) production and their respective receptors. Based on these results, the experimental conditions and receptors will be determined for blockade at the following assays. The role of PGE2 (EP1-4), endocanabinoid (CB1 and CB2) and angiotensin receptor 1 (AT1) receptors at the CPA function will be investigated by means of pharmacological blockade. The context of cellular differentiation and the envolvment of these inflammatory mediators at the inflammatory bone resorption mechanisms will be evaluated. (AU)
