Abstract

Salmonella Dublin is a serovar strongly adapted to cattle causing enteritis and/or systemic disease with high rates of mortality. However, sporadically it can be isolated from humans, usually causing serious disease, especially in patients with underlying chronic diseases. Differently of S. Dublin, bloodstream infections caused by other non-typhoidal serovars are not usual. However, in the Subsaarian Africa, non-typhoidal salmonella are among the most isolated pathogens related to bacteremia and meningitis. Salmonella Typhimurium accounts for a significant proportion of these infections and, strains belonging to a specific MultiLocus Sequence Type (MLST), the ST 313, have been related to these cases. A possible virulence marker found in the strains of the ST 313, the st313-td gene, interestingly is not present in the S. Typhimurium strains from other STs. However, it has been reported that this gene has been found in all S. Dublin strains. This project aim to verify the prevalence and expression of the st313-td gene, the expression of the genes sopE2 e fliC, related to invasion and flagella release, respectively, besides the ability to invade epithelial cells and survive in macrophages in Salmonella Dublin strains isolated in Brazil. For this, 115 S. Dublin strains isolated from humans and animals in Brazil will be studied. The st-313td gene will be searched by PCR and 20 strains will be selected for the human epithelial cell invasion assays (CACO-2) and survival in human macrophages (U937). Among these 20 strains, five will be selected to verify the expression of the st313-td, sopE2 and fliC genes. The results will provide data about the frequency and expression of the st313-td gene in S. Dublin strains isolated in Brazil. Moreover, this study will provide additional data about serovar Dublin, which few have studied, regarding the virulence mechanisms that permit it to cause invasive disease in humans and animals.
