Abstract

In this project, we propose to evaluate the ability of cationic nanoemulsions to promote cutaneous penetration and localization of the cytotoxic compound piplartine as a novel treatment option for tumors affecting the skin. The high incidence of skin tumors, the need of self-administered topical treatment options for patients for whom the surgery is not recommended, and the absence of topical formulations targeting the cutaneous localization of therapeutics agents for maximizing efficacy justify our proposal. Piplartine is an alkaloid / amide component of Piper species that has significant cytotoxic and antiproliferative activity in several tumor cell lines. As nanocarrier, we propose the use of oil-in-water cationic nanoemulsions with bioadhesive properties in order to increase the retention and the formulation interaction with the tissue. The formulations will be selected according to particle size, charge and rheological characteristics. Then, how these parameters influence the stability will be studied, as well as cutaneous permeability and the transport and localization of piplartine in/through the skin. The formulations cytotoxic effects will be evaluated in cutaneous equivalents of skin cancer. (AU)
