Physiological and biochemical mechanisms of thermal response to systemic inflammation in chicks

Abstract

During severe sepsis and associated systemic inflammatory responses, a drop in body temperature (Tc) instead fever can occur. Previously considered an uncontrolled response, nowadays it has been interpreted as a regulated, transitory and beneficial event in mammals. Although it has been described in birds, there is no evidence that this thermal response is regulated or not and no data of what are the specific mechanisms involved in the biphasic response (initial drop in Tc followed by fever) of systemic inflammation. Thus, the hypothesis of the present study is that the drop in Tc during systemic inflammation in birds is a regulated response, mediated by specific pro-inflammatory agents and represents a reduction of energy expenditure for the animal. To test this hypothesis, we will use a model of high dose of LPS (Gram negative bacteria endotoxin; 100 ¼g kg-1) peripherally injected in 5-day-old broiler chicks and analyze during biphasic response: i) autonomic (metabolic rate, pulmonary ventilation, cutaneous heat loss index) and behavioral (huddling) thermo-effector responses; ii) muscular and hepatic mitochondrial activities; iii) thermo-effector responses in a condition of reduced energy availability; iv) and the role of cyclooxygenases 1 and 2 as well as prostaglandins E2 and D2.
