Abstract

Bacterial resistance is one of the most important topics in collective health worldwide, characterized by the bacteria capacity to resist the effects of medication previously used to treat them. The rise of resistant bacteria and diseases dissemination increase more rapidly than the antibiotic development. To overcome this world crisis in the medical area, many strategies has been investigated, including the combined use of metallic nanoparticles and conventional antibiotics, to explore the synergistic effect of both antimicrobial agents. The development of nanocarriers emerges as a promising alternative, allowing high drug stability, high carrier capacity, feasibility of incorporation of both hydrophilic and hydrophobic substances, and feasibility of variable routes of administration, including intravenous application and inhalation. The main goal of this research project is to develop degradable acetalated dextran nanoparticles (Ac-DEX NPs) for tunable co-delivery of silver nanoparticles (AgNPs) and antibiotics. The project consists in four main stages: (I) use single and double emulsion methods to obtain Ac-DEX NPs containing hydrophilic (Izoniazid) and hydrophobic (Doxycycline) antibiotics, respectively; (II) the double emulsion method will be investigated to encapsulate the hydrophilic or hydrophobic antibiotic along with AgNPs; (III) AgNPs will be synthesized in situ, using D-glucose as the reducing agent and Ac-DEX as the capping polymer; (IV) the minimum inhibitory concentration (MIC) assay will be utilized to evaluate and compare the antimicrobial agents effects on bacterial viability (AU)
