Abstract

The current project of scientific initiation aims to evaluate the marking of mesenchymal stem cells (MSC) of the human bone marrow with nanoparticles that have magnetic properties and double fluorescent labeling, which enable them being detected by magnetic resonance imaging and by in vitro (excitation / emission: 555 / 579nm) and in vivo (absorption / emission: 755 / 777nm) fluorescence imaging techniques looking forward the traceability, migration and viability of MSC after administration in animal models. Another strategy that will be used for the tracing is bioluminescence, since the MSC will be transfected with luciferase. Besides that, it will allow to observe the viability of the MSC. The present subproject make part of a regular aid project FAPESP N ° 16 / 21470-3, entitled "Therapeutic action of mesenchymal stem cells of the human bone marrow, marked with multimodal nanoparticles, in diabetic rats submitted to focal cerebral ischemia: study of cellular, molecular and functional mechanisms". The project will be divided into five stages: the first step consist in the study of multimodal nanoparticles (NPMM) stability in culture under the conditions that will be used in the labeling of MSC. The second step corresponds to the marking of the MSC with the NPMM. The third step will analyze the intracellular distribution by visualizing light microscopy in the presence of iron and fluorescence in vitro, using one of the fluorophores (555 / 579nm) for the detection of NPMM in MSC. In the fourth stage, the in vitro evaluation through Magnetic Resonance Imaging (MRI) will be performed. This phase will be realized by Dr. Javier Bustamante Mamani and accompanied by the student of scientific initiation. In the fifth stage, the evaluation by fluorescence tomography in the infrared and bioluminescence signal in the CTM will be realized. This phase will be accompanied by Dr. Helio Rodrigues da Silva, research associate of the project.
