Abstract

Current trends in implant therapy have included modifying surfaces using nanotechnology tools and principles of bioengineering, increasing its performance when deployed. In addition, it is sought to know the biological responses involved in different alloys, still in tests, so that mechanical adjustments or even alterations in their physical-chemical properties, can adjust and improve the cellular response to these materials, thus predicting their performance When implanted in the hosts. Although much has been achieved in tools for the development of these materials, biological analysis methodologies have not advanced at this speed. The objective of this project is to validate the Ti-Sn-Nb alloy with Mg using osteoblasts derived from the bone marrow of equines and after, verify the biocompatibility of this framework against the bone fractures of rabbits. For this, initially, we will characterize the powders of the alloy, as well as their weighing and synthesis, through a planetary type mill. Next, we will compose the porous materials and their sintering in a resistive oven (1200 ° C). Afterwards, a topographic evaluation will be carried out by Electron Microscopy of X-ray Scanning and Diffraction, as well as by the Archimedes Test, to verify the apparent porosity and wettability of the material. Next, we will verify the cytotoxic effect of these materials, using a classic methodology for this purpose: Thiazolyl blue tetrazoluim bromide (MTT). Subsequently, through direct indirect methods, we will evaluate the biological response in vitro against these materials. In the indirect test, we will use seven experimental groups, with and without the conditioned medium of this alloy, (the specimens will be kept in culture medium for 24 hours), which will be used to treat semi-confluent osteoblasts. After 24 hours, the samples will be evaluated for cell migration, and after 35 days, the profile of cell differentiation and mineralization will be evaluated by Alizarin and Alizarin Red Phosphatase activity. On the other hand, in the direct contact tests, the osteoblasts will be cultured directly on the materials (10x104 cells / material) and, after 24 hours, the samples will be treated and taken for a morphological monitoring of these cells in contact with the materials through microscopy Scanning electron microscopy, with the purpose of knowing potential morphological variations in response to the materials under analysis. In addition, for the in vivo analysis, 7 New Zealand rabbits will be used, to which the frameworks will be implanted, together with their respective treatments in order to obtain the desired biological responses, and thus, we intend that the product generated serves as a parameter Of reference for new analyzes, offering new analysis methodologies, focusing its diffusion in the productive sector in the area of biomaterials.
