Abstract

Post breeding endometritis (EPPC) stands out as one of the leading causes of infertility in mares. In this context, the treatment with immunomodulators (IMM) has become popular as an option to increase the indices of conception in animals with EPPC. IMMs act directly on inflammatory mediators, blocking or attenuating an exacerbated or persistent inflammatory response, reducing post-coverage uterine inflammation. The medium conditioned by stem cells (MC), representing the supernatant obtained from the stem cell culture, where pre and inflammatory factors are released, as well as microvesicles with important role in tissue repair and in the control of oxidative stress. Because of its good, MC represents a new therapeutic option, with an immunomodulatory and tissue regenerative function. The objective of this study was to evaluate the effect of different concentrations of the environment over time and to analyze a modulation of the inflammatory response by physical and ultrasound examination and fertility evaluation. To carry out the project is direct 15 stallions and 20 mares with known reproductive history. For a better understanding of the analyzes of the project will be divided into two experiments. The project is divided into three phases. Phase 1 will be conducted in the laboratory with the objective of evaluating the effect of the addition of different concentrations of dog MC and equine on kinetics and sperm integrity, in order to determine the best conditioned medium / sperm quality ratio. In Experiment 2, the modulating capacity of the CM over the endometrial inflammatory process of mares submitted to artificial insemination will be evaluated in vivo, as well as the effect of the treatment of the semen with the conditioned medium on the conception rates of the equine females. In stage 3 MC will be used in mares with persistent endometritis after coverage. A statistical analysis of the curve fitting test (BioStat 4.0, Belém / PA, Brazil) was used to determine the best regression model to evaluate the effect of the explanatory variable represented by different concentrations of the environment on equine kinetics and integrity of equine spermatozoa. The same model will be used to determine the effects of MC treatment on the endometrial inflammatory response and fertility of equine females. Effects such as time of year, age and body condition score of mares, in addition to their possible interactions, will be evaluated through a general linear model (GLIMMIX, SAS Institute Inc, Cary, USA).
