Abstract

The formation of lipid rafts was initially proposed based on the preferential interactions between sphingolipids and cholesterol. While the basic Biophysics for the composition of lipid rafts is explained by simple principles, the theory for the selective incorporation of proteins into more ordered domains remains a mystery. Lipid rafts are usually located in regions of high concentration of proteins and lipids and have been associated with important biological processes. It is known that different lipid domains have different physicochemical properties, defined according to their packing and depending on the interactions between saturated lipids and cholesterol. Despite the importance of lipid rafts, little is known about the actual mechanism of its effect on cellular function. Thus, in this project we intend to study the lipid rafts and their interactions with biomolecules such as the enzyme cholesterol oxidase and the biopolymer chitosan. With the enzyme we shall study the influence of cholesterol on the membranes, since cholesterol oxidase is responsible for the oxidation of cholesterol. From the studies with chitosan, we expect to determine the permeation mechanism of the membrane, which is relevant for the use of chitosan as drug carrier. (AU)
