Abstract

Plant-pathogen interaction is a fundamental process that impacts agroecosystems. Understand the biological components and processes that control these interactions is expected to improve crop productivity. The thematic project: "Genes, Genomes & Transposable Elements contribution to plant-microbe interaction: a sugarcane study case" (FAPESP 2016/17545-8) aims to tackle this complex system using genome-wide molecular and bioinformatic tools to uncover key genes that control or mediate the interaction of sugarcane with three pathogens (Leifsonia xyli subsp. xyli; Xanthomonas albilineans; Sporisorium scitamineum). As part of the thematic project and to support the experimental designs proposed, this project have as main goals (i) the selection of sugarcane ROS genes and defense genes based in cell wall proteomics approach; (ii) the use of a real-time PCR assay to early detect and monitor the bacterial pathogens L. xyli subsp. xyli and X. albilineans and the sugarcane endosymbiont Gluconacetobacter diazotrophicus, to determine the time points to be used for the RNA capture experiments; (iii) validate sugarcane oxidative burst responses and pathogens protecting mechanisms against oxidative stress in sugarcane resistant and susceptible genotypes. We expect to identify key genes of the host defense machinery and microbial pathogenesis that could be used for screening resistant genotypes and to gain knowledge on plant-pathogen interactions.
