Abstract

Our laboratory has shown that the baroreflex was able to modulate local inflammatory responses, for example, in the joint, through sympathetic inhibition (Bassi et al., 2015), while the carotid chemoreflex was able to modulate the systemic inflammatory responses involving the autonomic sympathetic and parasympathetic nervous system (Santos - Almeida et al., 2017). On the other hand, periodontitis, a disease that affects 30-60% of the population, exhibits local (jaw) and systemic inflammatory characteristics (vascular dysfunction / arterial hypertension and inflammatory bowel diseases); at the present moment periodontitis does not present an effective therapy, except surgical approaches. Thus, motivated by clinical studies of carotid baroreflex activation for the treatment of resistant hypertension (Grassi et al., 2016), as well as unilateral carotid body resection (Narkiewicz et al., 2016), it is proposed in the present study to examine the role of electrical activation of the carotid sinus nerve, in conscious rat, with the purpose of characterizing whether the baro- and the chemoreflex are capable of modulating the local and/or systemic inflammatory responses, observed after ligature-induced periodontitis model of the jaw. Wistar Hannover rats will be submitted, under anaesthesia, to mandibular ligature for induction of periodontitis and implantation of a pair of electrodes around the carotid sinus nerve. Thus, each animal will have the carotid nerve chronically (09 days) stimulated every day at 17:00, with the following parameters: 0.5 mA; 30; 0.25 ms duration, for 15 minutes. On the eighth day after ligation of the jaw and implantation of the electrodes, the animals will have implanted, under anesthesia, a catheter into the femoral artery, in order to record blood pressure and heart rate on the following day; these records have the purpose to verify the efficacy of the electrical stimulation of the carotid nerve, by means of the hypotensive and bradycardic responses. Also, blood samples (1 mL) will be collected for analysis of plasma cytokines. Next, the animals will be anaesthetized for bone and gingival biopsies, and then submitted to decapitation, byguillotine, to collect the intestinal epithelium, which will be frozen for further analysis of tissue cytokines.
