Abstract

The social wasp Polybia paulista (Hymenoptera, Vespidae), abundant in southeastern Brazil, is extremely aggressive and responsible for many severe allergic reactions. One of the most allergenic components found in this venom is Antigen 5 (Poly p 5). Although the function of Antigen 5 has not yet been clarified, the molecule is a strong candidate to be used in immunotherapy for patients allergic to social wasp venom. Recently, it was demonstrated by our group that the soluble recombinant form of Poly p 5 (ryPoly p 5) obtained through expression in Komagataella phaffii is allergenic and induces an immune response that is comparable to its native counterpart. As a prerequisite to use ryPoly p 5 for CRD (Component-resolved diagnostics) of P. paulista venom allergy it has to be characterized in detail on a molecular and immunological level. For this characterization, cross-reactivity analyses with crude venom from temperate species are of crucial importance. This is due to the fact that the main diagnostic systems available on the market are based on recombinant allergens of species from the Northern hemisphere. Therefore, in this work we aim at the characterization of the immunological potential of recombinant antigen 5 from Polybia paulista as a prerequisite to use this allergen in the routine diagnosis of allergies to the venom of social Hymenoptera. Moreover, the obtained results could contribute to the understanding of the biological role of the allergen Antigen 5 in Hymenoptera venoms from the region of Europe as well as from the region of South America.
