Allergens from venom of the social wasp Polybia paulista (Hymenoptera, Vespidae): recombinant expression in yeast for improved diagnosis and specific immunotherapy to Hymenoptera venom allergy

Abstract

The social wasp Polybia paulista (Hymenoptera, Vespidae) is quite abundant and endemic in the states of São Paulo and southern Minas Gerais. It has urban habits and is very aggressive, causing many significant accidents of medical perspective. To date, there are no allergenic extracts or components available in our country to diagnosis and treatment of allergic patients to insect venom despite the great number of existing species. Through biochemical, immunological analysis and of IgE recognition in the serum of allergic patients to venom of this wasp, we aimed to perform a comparative analysis of the immunogenic potential of the recombinant allergens (phospholipase A, antigen 5 and hyaluronidase) expressed in Pichia pastoris, in comparison with the venom allergenic native proteins. Our results will contribute to advances in the treatment of specific immunotherapy (IT) and to develop diagnostic kits for allergies caused by the venom of P. paulista and of other closely related species. Taking in account that these are the first recombinant venom allergens from a Brazilian endemic wasp species, this comes to fill a shortage in Brazil since today our patients are treated with wasp venom extracts from temperate countries.
