Abstract

The excessive accumulation of body fat produces several harmful effects on human body. One of those effects is the metabolic disorder, which is associated to the begging of the insulin resistance and the diabetes mellitus type 2, which is defined as hyperglycemia and an impairment in insulin response and secretion. The diabetes induction by high fat diet is well known and commonly used because it is similar to the human diet. The low-level laser therapy (LLLT) induces the increase of cellular metabolism, regeneration, proliferation and protein synthesis. Thus, it modulates the inflammatory process and controls pain. However, very little is known about the effects of the LLLT on the progress and control of the diabetes. Putting together, the aim of this study is to analyse the effects of the LLLT on the glucose homeostasis and the insulin expression in pancreas of mice fed with a high fat diet for 12 weeks. Our hypothesis is that the LLLT will minimize the malefic effects of the high fat diet, slowing the progress of diabetes. We will be used C57bl6 mice, housed in the Central Bioterium of the Federal University of São Paulo - Baixada Santista, with a 12:12-h light-dark cycle, temperature controlled 22ºC ± 2ºC, food and water ad libitum. The mice will be divided in 3 different groups, which two of them will be fed with a high fat diet: control group (C), high fat diet group (HDF) and high fat diet + LLLT (HFD + LLLT). The LLLT utilized will be the GalAlAs, presenting 808 nm wavelength, continuous emission, 100 mW potential outflow, 0.028 cm2 light beam, 3.57 W/cm² irradiance, 30 J/cm² dose, 0.84 pontual irradiation energy and irradiation time of 8 seconds. Body weight and food intake will be verified weekly. At the end of the experimental time, intraperitoneal tests will be performed to verify the glucose and insulin tolerance. The fast blood glucose will be also determined. After the euthanasia by decapitation, morphometric analyses of the endocrine pancreas will be realized such as the immunoperoxidase for insulin detection. For statistical analysis, normality of variables will be verified to determine the most appropriate hypotheses test, using SPSS software version 20. Significance will be set at 95%.
