Abstract

Chikungunya virus (CHIKV) causes acute and often symptomatic infections that are associated with fever and debilitating polyarthralgia/polyarthritis. CHIKV infection has also been associated with high lethality among elderly patients with preexisting conditions. Surprisingly, over 25% of CHIKV-related deaths in the Americas have been reported in Fortaleza city, Ceará State, Brazil. In this context, this project aims to understand the evolutionary and pathogenesis mechanisms associated with CHIKV-related deaths. To achieve this, we will perform a multi-platform omics analysis of peripheral samples recovered from different body compartments of fatal and non-fatal CHIKV patients to identify key virus-host interactions and biomarkers underlying disease severity. We will conduct extensive studies of CHIKV genetic diversity across viral genomes to identify selective pressures and hotspots of virus diversity along the genome that may be associated with disease severity. Finally, we will create a data-centric bioinformatic environment for the visualization of genomic, clinical and epidemiological data related to arbovirus circulation in Brazil. This study will shed light into the mechanisms underlying CHIKV severity, and will provide a valuable community resource for the analysis of genomic, clinical and epidemiological data on arboviruses circulating in Brazil.
