Abstract

MicroRNA are non-coding single stranded RNAs that can act autocrine, paracrine and endocrine to regulate metabolic pathways related to the development of Noncommunicable Diseases (NCDs). In addition, microRNA act as potential NCDs biomarkers, since the plasma expression pattern of microRNA can be altered according to the individual's health and lifestyle. Given the above, the present project aims to evaluate the expression profile of microRNA in plasma and to verify its relationship with cardiometabolic biomarkers and diet in adult individuals participating in a population-based study. For this, a cross-sectional study will be developed, with a subsample of 200 adult individuals, aged 20 to 59 years old, participating in the Isa-capital study conducted in São Paulo, in 2015. The plasma expression profile of 22 microRNAs related to glycemic and lipid metabolism, adiposity and inflammatory response will be evaluated. Together, inflammatory biomarkers will be determined [interleukin (IL) -1², IL-6, IL-10, tumor necrosis factor alpha, monocyte chemotactic protein 1, plasminogen activator inhibitor 1, C-reactive protein, adiponectin, leptin, and soluble intercellular 1 and vascular adhesion molecules 1]. Data will also be used regarding anthropometric measurements (weight, height and waist circumference); systemic blood pressure; diet (two 24-hour recalls); and glycemic (fasting blood glucose and plasma insulin) and lipid (cholesterol, LDL-c, HDL-c and VLDL-c and triacylglycerol) markers of these individuals. Results will be expressed as measures of central tendency and dispersion or as a percentage and 95% confidence interval. Data distribution will be verified by Kolmogorov Smirnov test and subsequent tests chosen accordingly. The relationship between the variables of interest will be verified by multivariate analysis. Values of p <0.05 will be considered statistically significant. With the execution of the project we seek to expand the knowledge about the use of microRNA as biomarkers for NCDs, thus improving the risk reduction and treatment processes of these diseases. (AU)
