Abstract

Periodontitis is the one of the most prevalent diseases in the world. This disease is driven by and propagated by a pathogenic plaque biofilm and characterized by a dysregulated and excessive inflammatory response. Natural botanical substances have been widely shown to have anti-inflammatory and antimicrobial properties, and therefore have potential for use in the management of periodontitis. However, one of the key challenges in using these natural substances is their limited stability, are easily degraded and present low aqueous solubility limiting therapeutic use. To mitigate this problem, we have developed multiparticulate mucoadhesive systems, for sustained release of the natural substances (morin and carvacrol) thereby enhancing their therapeutic potential. This research project aims to evaluate the anti-inflammatory, antioxidant and antimicrobial activity of these mucoadhesive systems, against periodontal pathogens. This will utilize a well-established H400 oral keratinocytes cell line model of periodontal inflammation which will be exposed to periodontal pathogenic bacteria (P. gingivalis and F. nucleatum) for 24 h. Cells will be pretreated with the chosen system alongside non-active controls. Following incubation cell media with be tested inflammatory cytokines by ELISA. Gene expression levels of inflammasomes NLRP3 and IL-1² will be evaluated by RT - PCR and western blot. For antioxidant capacity neutrophils isolated from the peripheral venous blood of periodontally and systemically healthy volunteers will be stimulated by non-viable P. gingivalis and F. nucleatum and the ability of the chosen system to reduce extracellular and total reactive oxygen species production determined. For antimicrobial evaluation P. gingivalis, and F. nucleatum will be treated with the system in both biofilm and planktonic culture. The inhibitory effect of the system on bacterial planktonic cultures will be performed both turbidity assessment and colony forming unit (CFU). A live/dead staining assay and CFU will be utilized to determine the effect of the system on bacterial viability in mature biofilms. (AU)
