Abstract

Introduction: Dietary and surgical treatments for obesity can modulate epigeneticregulation, acting on energy metabolism and altering the transcription of genesunderlying oxidative stress, inflammation, and metabolic imbalance. Objective: tocomparatively analyze the methylation profile of genes related to energy metabolism inpatients before and after bariatric surgery. Methodology: individuals undergoing Rouxen-Y gastric bypass (RYGB) will be selected before and after six months of surgery.Anthropometric indicators, food consumption, biochemical indicators and calorimetrywill be evaluated. In addition, whole blood DNA will be extracted for DNA methylationanalysis, which will be performed using the Infinium Methylation EPIC Kit. The datawill be analyzed by the ChAMP package (Chip Analysis Methylation Pipeline), whichwill select the differentially methylated regions (DMRs). After processing, the resultingdata will be enriched in the STRING (Protein-Protein Interaction Networks FunctionalEnrichment Analysis) platform for gene ontology results. For descriptive statistics,mean and standard deviation will be used. The Shapiro-Wilk test will be used to verifythe normality of the data. Afterwards, the following will be applied: paired t-test or thecorresponding non-parametric test, Wilcoxon test, Pearson/Spearman correlation andlinear regressions to evaluate the differences between the variables in the pre and postbariatric surgery periods. The admitted significance level will be p<0.05.
